Le Antioxidant Activator (NF-E2-related factor 2), un facteur de transcription humain codé par le gène NFE2L2, régule l'expression d'un ensemble de gènes antioxydants et détoxifiants. En l'absence de stress, Nrf2 est ancré dans le cytoplasme par son inhibiteur spécifique Keap1 (kelch-like ECH-associated protein 1). Keap1 fonctionne comme un capteur d'oxydants et de xénobiotiques électrophiles. En présence de l'une de ces substances, Keap1 cesse d'inhiber Nrf2. Cette action stabilise Nrf2, lui permettant de s'accumuler dans le noyau et de se lier à l'élément de réponse antioxydant (ARE) situé dans les enhancers de ses gènes cibles. Dans ces conditions, Nrf2 régule alors à la hausse une variété d'enzymes antioxydantes et de protéines détoxifiantes.
Source of antioxidants for the maintenance of good health.Nrf2 (NF-E2-related factor 2), a human transcription factor encoded by the NFE2L2 gene, regulates the expression of a set of antioxidant and detoxifying genes. In the absence of stress, Nrf2 is anchored in the cytoplasm by its specific inhibitor Keap1 (kelch-like ECH-associated protein 1). Keap1 functions as a sensor for electrophilic oxidants and xenobiotics. In the presence of either of these substances, Keap1 ceases to inhibit Nrf2. This action stabilizes Nrf2, allowing it to accumulate in the nucleus and bind to the antioxidant response element (ARE) located in the enhancers of its target genes. Under these conditions, Nrf2 then upregulates a variety of antioxidant enzymes and detoxifying proteins.
